Using the mineral salt medium allowed the cells to be grown from the beginning in order to eliminate complicated steps with other protocols (incubating the cells in LB medium, followed by washing, and further incubating in the depleted medium), meaning that photo-methionine could be incorporated at the very beginning of the cell growth that had a high yield above 30%. Photo-methionine had shown no damage during the cell growth process and once photo-activated by UV-A light, CaM had nine distinct cross-link sites once MS had determined there was peaks of photo-methionine labeled CaM.
